黑曲霉代谢组分对芝麻幼苗的化感作用研究

从黑曲霉产物对芝麻种子的发芽影响和薄层检测结果可以看出,组分6和组分7抑制最强,但没有明显的显色斑点,其样品质量分别仅有1mg和1.25mg。


摘  要:本实验研究了加拿大一枝黄花内生黑曲霉发酵产物对芝麻种子发芽及幼苗生长的影响。以培养皿滤纸培养法,用不同分离组段分别处理种子后,检测各组发芽率、发芽指数及幼苗生物量和长度变化,确定发酵产物的乙酸乙酯洗脱段(组段2)具有良好的化感活性,该处理组中芝麻的平均发芽率为27.38%,对照组为42.22%。通过凝胶柱层析分离和高效液相色谱(HPLC)检测,获得具有单一成分的洗脱组段C1,并检测C1对芝麻幼苗生长特性的影响。结果表明不同浓度的C1对芝麻幼苗的芽长、叶绿素a和类胡萝卜素含量都有较强的抑制作用,低浓度和高浓度处理组的芽长分别为水对照组的85%和81%,叶绿素a均为对照组的57%,类胡萝卜素分别为对照组的83%和121%。C1对芝麻幼苗根长则表现出很强的促进作用,C1处理后芝麻的根长可分别达到水对照组的305%和236%。对芝麻幼苗中的叶绿素b含量有轻微促进作用,高低浓度组的含量分别为水对照组的104%和190%。C1对芝麻幼苗中的蛋白质含量和多糖含量影响较弱,两种处理组的多糖含量分别为水对照组的120%和98%,蛋白质含量分别为水对照组的112%和81%,表现出低促高抑的作用特点。

Abstract:In this experiment, the effects of fermentation products of Aspergillus sp. on the germination and seedling growth of sesame seeds were studied. The germination rate, germination index and seedling biomass and length were determined by using different culture fractions. The ethyl acetate elution section (fraction 2) of the fermentation product was determined to be good The average germination rate of sesame seeds in this treatment group was 27.38%,The control fraction was 42.22%.The elution fraction C1 with single component was obtained by gel column chromatography and high performance liquid chromatography (HPLC), and the effect of C1 on the growth characteristics of sesame seedling was tested. The results showed that the buds of different concentrations of C1 had strong inhibitory effects on the bud length, chlorophyll a and carotenoid contents of sesame seedling. The buds of low concentration and high concentration group were 85% and 81% of water control group, Chlorophyll a was 57% of the control group, carotenoids were 83% and 121% of the control group, respectively. C1 showed a strong promoting effect on the root length of sesame seedling. The root length of sesame seeds could reach 305% and 236% of the water control group respectively.
