摘要:使用D-半乳糖注射诱导衰老模型,探究葡萄籽原花青素提取物对衰老模型小鼠肠道菌群结构的影响。将C57BL小鼠分成随机的对照组、衰老组和葡萄籽原花青素干预组,6周后测定肠组织中T-AOC、SOD、CAT和MDA含量,利用Illumina MiSeq高通量测序技术来探究衰老小鼠肠道菌群的影响,并利用GC分析技术探究肠道菌群代谢产物短链脂肪酸含量的影响。结果表明,衰老组小鼠抗氧化酶活性明显降低,MDA含量明显增长。在葡萄籽原花青素干预后,小鼠体内的多种抗氧化酶活性上升,MDA含量显著降低,与对照组没有显著差异。三组肠道菌群多样性无显著差别。但在门水平上,对照组与干预组中Bacteroides的相对丰度为显著高于衰老组,而Firmicutes的相对丰度则显著低于衰老组。在属水平上,Bacteroides-s24-7、Bacteroides、Blautia、Lachnospiraeae、Ruminiclostridium、Lachnospiraeae- NK4A136、Roseburia、Bilophlia等7种菌群的相对丰度存在显著性差异。相比对照组,衰老组的Bacteroides-s24-7的含量显著降低,其他菌种的含量是显著上升。与衰老组相比,在葡萄籽原花青素组中Bacteroides-s24-7, Roseburia、Lachnospiraeae-NK4A136和Bacteroides的含量是显著上升的,而Blautia、Lachnospiraeae、Ruminiclostridium和Bilophlia的含量是显著降低的。此外,原花青素的干预可显著增加小鼠粪便中代谢产物短链脂肪酸—乙酸、丙酸、丁酸含量。本研究表明原花青素增强衰老小鼠肠道抗氧化活性,调节肠道菌群结构,一定程度上增强产生丁酸类的有益菌相对丰度,提高短链脂肪酸的含量,进而延缓衰老进程。

The effects of procyanidins on regulation of intestinal microecological in D-galactose induced mice.
Abstract:The study is to investigate the effects of procyanidins B2 on intestinal microflora in induced aged mice with D-galactose injection. Thirty C57/BL mice were randomly pided into control group, aged group and the group treated with procyanidins B2. After seven weeks, we detected the activity of T-AOC, SOD and CAT and the content of MDA in the duodenum and colon. Using the Illumina MiSeq high-throughput sequencing technologies,we detected intestinal flora 16s rDNA gene to explore the relationship between the procyanidin B2 and intestinal flora in the aged mice.The result showed that activity of antioxidant enzyme was decreased in the two tissues of aged mice, the content of MDA was significantly increased. After the intervention of procyanidins B2, the activity of antioxidant enzymes in the aged mice was increased, and the content of MDA was decreased significantly. The persity of intestinal biopersity of aged mice was decreased significantly, and the persity was increased significantly after the intervention of procyanidins B2.Compared with three group,Bacteroides-s24-7,Bacteroides,Blautia,Lachnospiraeae,Ruminiclostridium,Lachnospiraeae-NK4A136, Roseburia, Bilophlia were changed significantly.Compared with the control group, the proportion of Bacteroides-s24-7 in the aged group was decreased significantly, and the proportion of other strains were increased significantly. Compared with the aged group, the proportion of Bacteroides-s24-7, Roseburia,Lachnospiraeae-NK4A136 and Bacteroides were significantly increased in the mice treated with procyanidins B2, while the proportion of Blautia, Lachnospiraeae, Ruminiclostridium and Bilophlia were significantly decreased.
