植物精油微胶囊对白色念珠菌的气相抑菌研究

植物精油具有抑菌效果已获得广泛认可,但如何提高植物精油的抑菌效率,这是一个至今仍旧困扰研究人员的问题。


摘要:植物精油是一种提取自天然植物的安全抑菌物质,因其抑菌效果优良且副作用小而被广泛关注。微囊化可用来解决精油易挥发、对光热敏感等问题,该方法用安全的复合壁材包埋植物精油,有效的拓宽了植物精油的商业市场。白色念珠菌(Candida albicans)是一种常见的致病微生物,可运用植物精油微胶囊抑制其在食品中的生长,进而延长食品货架期。本文熏蒸植物精油微胶囊,以研究微胶囊技术及其主要成分的气相抑菌效果。主要研究成果如下:(1)通过改良的滤纸片熏蒸方法,研究3种植物精油及其主要成分微胶囊对白色念珠菌的气相抑制效果,发现抑菌圈明显随微胶囊浓度增加而增大,综合考虑风味的接受程度,选择香茅及香茅醇微胶囊用于后续气相抑制白色念珠菌的规律研究;(2)研究28和40℃条件下,纯精油及其微胶囊挥发0、1、3和5 d后的气相抑菌效果,结果显示香茅精油、香茅醇及其微胶囊的抑菌性能均与挥发时间成反比,挥发时间越长,抑菌圈越小,但在40℃时,香茅精油和香茅醇对比等量精油含量的微胶囊,在挥发7d后的抑菌圈显著小于微胶囊处理;(3)pH对香茅精油和香茅醇微胶囊气相抑菌效果影响研究显示,当培养基为4、5.5、7和8.5时,纯精油熏蒸的抑菌圈无明显影响,但香茅精油微胶囊的气相抑菌性会受pH影响,随着pH的增加而明显降低。(4)将含有精油及其微胶囊的滤纸片悬挂于含有液体培养基的三角瓶上方,研究气相熏蒸对白色念珠菌在液体培养基中生长的影响,结果显示香茅精油及香茅精油微胶囊的OD值在0~24 h呈上升趋势,在24~48 h内呈下降趋势。进一步通过细胞通透性试验,发现香茅精油及香茅精油微胶囊对白色念珠菌细胞膜破坏的程度随时间的增大而增大;香茅精油在第0~32 h时,对白色念珠菌细胞膜破坏程度强于香茅精油微胶囊对白色念珠菌细胞膜破坏程度;在40~48 h,香茅精油对白色念珠菌细胞膜的破坏程度弱于香茅精油微胶囊对白色念珠菌细胞膜破坏程度。

Study on microcapsules of plant essential oil against candida albicans by air - phase inhibition

Abstract: Plant essential oil is a safe bacteriostatic substance extracted from natural plants.Microencapsulation can be used to solve such problems as volatile oil and sensitive to light and heat. This method USES safe composite wall materials to bury plant essential oil, effectively expanding the commercial market of plant essential oil.Candida albicans is a pathogenic microorganism commonly found in the skin and mouth. It can use microencapsulated plant essential oil to prevent it from contaminating food, thus extending the shelf life of food.In this paper, the effects of microcapsule technology and its main components on bacteriostatic effect were studied by air - phase bacteriostatic method.Main research results are as follows: (1) by the method of improved filter fumigation, studies three kinds of plant essential oil and its main ingredients microcapsules gas phase inhibition effect of candida albicans, bacteriostatic circle found significantly increased with the increase of concentration of microcapsule, comprehensive consider the acceptance of flavor, citronella and geraniol microcapsule used for subsequent law and research of gas phase inhibition of candida albicans,28 (2) research and under the conditions of 40 ℃, pure essential oils and microcapsule volatile 0, 1, 3, and 5 d after the bacteriostatic effect of gas phase, the results showed citronella oil, geraniol and antibacterial properties of the microcapsule is inversely proportional to the volatile time, volatilize the longer the time, the smaller the bacteriostatic circle, but in 40 ℃, citronella essence oil and geraniol contrast amount of essential oil content of microcapsules, bacteriostatic circle in volatile after 7 d significantly less than microcapsules handling;(3) the pH of citronella essence oil and geraniol microcapsule bacteriostatic effects study shows that the gas phase when the medium is 4, 5.5, and 8.5 7, pure essential oil fumigation of bacteriostatic ring has no obvious effect, but the citronella oil microcapsule bacteriostatic sex will be affected by the pH of the gas phase increased with the increase of pH is decreased obviously.(4) will contain essential oil and filter of microcapsule suspension in the triangle flask containing liquid medium above, study the gas phase fumigation on candida albicans in liquid medium growth, results show that the citronella oil and citronella oil microcapsule OD value in 0 ~ 24 h is on the rise, a downward trend in 24 ~ 48 h.Furthermore, the cell permeability test showed that the cell membrane damage of citronella oil and citronella oil microcapsules increased with time.When citronella essential oil was at 0~32 h, the cell membrane of candida albicans was damaged more than that of citronella microcapsule.At 40~48 h, the damage degree of citronella essential oil on the cell membrane of candida albicans was weaker than that of citronella essential oil microcapsule on candida albicans cell membrane.
