摘要:在当下,对于秀丽隐杆线虫的探索受到了越来越多国内外学者关注,秀丽隐杆线虫已然成为一种重要模型在各个探索领域中被人们经常使用。其具有研究周期短、突变体多的优点。本实验经过对猴头菇进行加蒸馏水、提取、过滤、浓缩、离心、醇洗、溶解、透析得到浓度为0mg/L、5mg/L、50 mg/L、500 mg/L的β葡聚糖。利用秀丽隐杆线虫为一种试验手段,来验证猴头菇β葡聚糖对线虫的抗氧化和抗衰老的影响,在实验中主要进行了线虫的寿命试验、抗氧化应激试验、线虫生殖试验、体外抗氧化DPPH试验、线虫运动能力试验等结果表明,较高浓度的β葡聚糖可以在某些程度上可以延长线虫的寿命。这些试验的尝试对于往后猴头菇β葡聚糖的研究应用中必定会有一定的指导意义。试验结果表明,高浓度的猴头菇β葡聚糖能够显著提高线虫的寿命,最适宜的浓度在500mg/L,但需要牺牲生殖率来做到。

Abstract:In the present time, the exploration of Caenorhabditis elegans has attracted more and more attention from scholars at home and abroad. Caenorhabditis elegans has become an important model that is often used by people in various fields of exploration. It has the advantages of short research period and many mutants.In this experiment, the Hericium erinaceus was added with distilled water, extracted, filtered, concentrated, centrifuged, washed with alcohol, dissolved and dialyzed to obtain β-glucan with concentrations of 0, 5, 50, 500 mg/L.The use of Caenorhabditis elegans as an experimental method to verify the effect of Hericium erinaceus β-glucan on the anti-oxidation and anti-aging of nematodes. In the experiment, the life test of nematodes, anti-oxidative stress test, and nematode reproduction were mainly carried out. Tests, in vitro antioxidant DPPH tests, and nematode exercise ability tests have shown that higher concentrations of beta glucan can extend the lifespan of nematodes to some extent. The trials of these experiments will surely have certain guiding significance for the research and application of the future Hericium erinaceus β-glucan. The test results showed that the high concentration of beta-glucan from Hericium erinaceus can significantly increase the lifespan of nematodes.The optimum concentration is 500 mg/L,but it is necessary to sacrifice the reproductive rate.
